You can't really lose targeted weight in your butt...just like you can't gain weight just in your butt. You can lose weight all over though.
The way to lose weight is through healthy diet and exercise. Eat 4-6 SMALL meals a day, consisting of lean meats, whole-grain starches, fruits and vegetables. Set up your plate so 1/4 is meat, 1/4 is starch, 1/2 is fruit and veggies, and there is minimal fat.
If you do this as well as drink 8 glasses of water a day and exercise for 30 min a day a few times a week, you will see healthy results.

Agreeing with what prkswllflwr said, I'm going to add on by telling you that pilates are actually a really good way to lose weight from the area or so I've heard, personally I've never worried about it but everyone that I know who has done them, has seen weight loss in their butt although they weren't exactly happy about it. =P btw pilates are a group of exercises that tone the butt, abdomin, and thighs if I'm not mistaken so doing a lot of these may very well give you weight loss results in these areas as well.
